چکیده

C-aryl derivatives of DOTA (N,N’,N”,N”’-tetrakis(carboxymethyl)1,4,7,10-tetraazacyclododecane) including phenyl-DOTA and 3-NO2Ph-DOTA were synthesized by a template reductive reaction. GaCl3 or GaCl3 was taken and transformed into Ga-acetylacetone (Ga(acac)3) before labeling to the DOTA derivatives. The Ga or Ga labeled complexes were generated by ligand-ligand exchange of acetylacetonate with the DOTA derivatives. The labeling efficiency of PhDOTA with Ga could be higher than 95% under the reaction being facilitated via sonication, at ambient temperature for 20 min. By a similar reaction, the labeling efficiency of 3-NO2-Ph-DOTA with Ga was found to be lower (~ 40%). The stability of Ga-Ph-DOTA was high, maintaining at ≥ 98% yield even at 24 h post-preparation. The Ga labeled Ph-DOTA complex was radiochemically characterized to be anionic and hydrophilic. It is anticipated from the result that Ga in the form of Ga(III) may be six coordinated with Ph-DOTA by 4N from the ring and 2O from two of four pendent acetic groups, leaving other two acetic groups uncoordinated.

Production and Quality Control of [67Ga]-DOTA-trastuzumab for Radioimmunoscintigraphy

Breast cancer radioimmunoscintigraphy targeting HER2/neu expression is a growing field of work in nuclear medicine research. In this study, trastuzumab was successively labeled with [(67)Ga] GaCl3 after conjugation with DOTA-NHS-ester. The conjugates were purified by molecular filtration, the average number of DOTA conjugated per mAb was calculated and total concentration was determined by spec...

Comparative studies of three 68Ga-labeled [Des-Arg10]kallidin derivatives for imaging bradykinin B1 receptor expression with PET.

UNLABELLED Bradykinin B1 receptor (B1R) is a G-protein-coupled receptor that is overexpressed in a variety of cancers. B1R is not expressed in healthy tissues, making it an attractive cancer imaging marker. Previously, we reported selective uptake of (68)Ga-P03034 ((68)Ga-DOTA-dPEG2-Lys-Arg-Pro-Hyp-Gly-Cha-Ser-Pro-Leu) in B1R-positive (B1R+) HEK293T::hB1R tumor xenografts in mice. In this study...

Development of Novel Radiogallium-Labeled Bone Imaging Agents Using Oligo-Aspartic Acid Peptides as Carriers

(68)Ga (T 1/2 = 68 min, a generator-produced nuclide) has great potential as a radionuclide for clinical positron emission tomography (PET). Because poly-glutamic and poly-aspartic acids have high affinity for hydroxyapatite, to develop new bone targeting (68)Ga-labeled bone imaging agents for PET, we used 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA) as a chelating site and c...

The Synthesis and Evaluations of the 68Ga-Lissamine Rhodamine B (LRB) as a New Radiotracer for Imaging Tumors by Positron Emission Tomography

PURPOSE The aim of this study is to synthesize and evaluate (68)Ga-labeled Lissamine Rhodamine B (LRB) as a new radiotracer for imaging MDA-MB-231 and MCF-7 cells induced tumor mice by positron emission tomography (PET). METHODS Firstly, we performed the radio synthesis and microPET imaging of (68)Ga(DOTA-LRB) in athymic nude mice bearing MDA-MB-231 and MCF-7 human breast cancer xenografts. A...

Influence of DOTA chelators on radiochemical purity and biodistribution in xenografted mice of 177Lu- and 90Y-Rituximab

Introduction This work presents a comparative biological evaluation of 90Y- and 177Lu- labelled DOTA-SCN and DOTA-NHS conjugated to Rituximab in tumour-bearing mice. Materials and methods Two DOTA derivatives, p-SCN-Bn-DOTA and DOTA-NHS-ester were conjugated to Rituximab and then freeze-dried kit formulations were prepared, as previously described [1]. Tissue distribution was investigated in t...
